Project 1: Glyco Insight into the Factor VIII Inhibitors of Hemophilia A Patients

Objectives

  • To develop glycoanalysis toolsets for in-depth study of factor VIII glycosylation and its inhibitors.
  • To survey novel glyco biosignatures related to FVIII inhibitor development in hemophilia patients.
  • To investigate the glycosylation changes in hemophilia A patients undergoing immune tolerance induction.

Project 2: Genetic Characterization of Factor VIII Inhibitors and Glycosylation Patterns

Objectives

  • To investigate the global effects of enhanced glycoforms/glycopeptides on factor VIII inhibitor formation.
  • To investigate the role of host glycosylation genetic environment on factor VIII inhibitor formation.
  • To investigate tissue/organ specific FVIII glycosylation and inhibitor formation.

Project 3: Immune Regulation by Cellular Glycosylation for the Inhibitory Antibody Development to Factor VIII

Objectives

  • To identify the key glycan profile of factor VIII inhibitors in selected hemophilia patients and to characterize the sensitivity of factor VIII inhibitors to FVIII-glycopeptides.
  • To examine the specificity of B and T cell glycopeptide epitopes identified by glyco array in mice.
  • To examine the influence of glycan profiles of various strains of mice on the development of anti-FVIII immune responses.
  • To examine the correlation of anti-factor VIII antibody production with T and B cell responses in hemophilia A mice following delivery of factor VIII proteins including FVIII and FVIII derivatives with different extent of glycosylation as well as FVIII variants with engineered glycosylation patterns.
